other than the obvious because i'm sure that we automatically can tell a boy for a boy and a girl for a girl. i hope.lol personally, i've recieved a comment of whether or not i look like a boy because of my hair. its midlength, so from the back one could assume that i'm a guy i guess, despite my huge rear end. anyways, i notice eye contact and expressions, and voice first. the voice is a really important part because you can learn a bit about a person's personality from their voice. for example if they speak mummbly and unenthusiastic that could mean the person is apathetic and slovenly. Loud and hard could mean they're a little stubborn and attention seekers. that last one is something i'm trying to fix.lol
I always notice if they have a certain accent or way of speaking. I also notice if they are the hugging type or the personal- space- issues type. ( I'm the hugging type and I have all space- issues friends! I need a hug)!
The way they smell and act. I have a very strong sense of smell and I kind of remember people by the way the smell. I hope that doesn't make me sound like a creeper as if I'm getting on people and smelling them...but it's just when I sit beside someone they have a certain personal smell and that's usually the first thing I notice. If I can't notice that they way they act or look comes second. Like I check to see if they're slouching, if they're hygienic, etc. or if they act shy or over-confident.

I'm kind of sensitive to smell too, but that's even before I look. Like how I smelt the cheap perfume of my excessively lewd male teacher and instinctively backed off.
First, whether their wearing glasses.
Second, their height.
Third, their hair.
When I take a gander at someone, it is hard to tell what I will notice first, other than the gender and race of course. I pay attention to anything that sticks out to me, if that is their humngous nose, than there I am focusing on their nose. If it is their eyes though, I will look at them, but then I will look away. For some odd reason, when I take to someone, I cannot actually look them in the eyes.
