Something that really helps is to have at least one complete draft of the story. Once you have an idea of what needs to happen, it's a lot easier to figure out the hows and whys, which is where the characters come in.

For example, why is a character seeking, say, a mystical stone of legendary powers? How does the character intend to acquire it? What lengths are they willing to go to to get the stone? How will they react to obstacles in their way, people who interfere, events that delay them, etc.?

Once you have a character's motivations pinned down, a lot of the other characterization things will fall into place, at least from a plot perspective.

@Kyllorac I tried what you suggested and it's working really well. I still need to tweak a few things with my character's personality but I think its working out. Thanks for your awesome advice.
